In this work, we demonstrate the complete microfluidic preparation of amplifiable virus DNA from dilute specimens on a single device. For the first time, free-flow electrophoresis is used to concentrate the viral sample followed by thermal lysis and gel electrophoretic nucleic acid extraction. The small size of the chip, the user-friendly operation with low risk of cross-contamination and the high potential for automation, makes it an ideal candidate in the field of point-of-care diagnostics.
